Exemestane is steroidal aromatase inactivator. Its structure is similar to the natural substrate of aromatase, andolene two ketone, which is the pseudometallic substrate of aromatase. Since the estrogen of postmenopausal women is mainly transformed by androgens (adrenocortical production) under the action of aromatase in the peripheral tissue, this product inactivated by the irreversible binding of aromatase active sites, thus reducing the level of estrogen in the blood circulation of menopause women. This product has no obvious effect on the biosynthesis of corticosteroids, and there is no significant effect on other enzymes in the corticosteroid generation pathway even if the concentration is 600 times higher than the inhibitory concentration of aromatase.
Oral absorption of this product is rapid, and absorption is affected by food. Oral bioavailability is 42%. After menopause, the absorption rate of the drug was higher than that of the healthy subjects.
