Tacrolimus is a macrolide isolated from Streptomyces genus, which has a strong immunosuppressive effect. It can specifically bind and inhibit the activity of calmodulin phosphatase, inhibit the transcription of il-2 signal, and thereby inhibit the activation of T cells, the production of TNF-, il-1 and il-6, and the proliferation of T cell-dependent B cells. Compared with cyclosporine, another calmodulin phosphatase inhibitor, tacrolimus has a stronger inhibitory effect on T cell activation with fewer adverse reactions.
